ردیابی گونه های فوزاریوم مولد مایکوتوکسین با استفاده از صفات ریخت شناسی و مولکولی در دانه ذرت

چکیده

چکیده
قارچ فوزاریوم از بیمارگرهای عمده­ی مزارع ذرت محسوب می­شود که علاوه بر تقلیل کمیت و کیفیت محصول، با تولید چند نوع مایکوتوکسین از جمله فومونیزین (FUM)، موجب آلودگی مواد غذایی و تهدید سلامت انسان و دام می­شود. در تحقیق حاضر نمونه­برداری از دانه­های ذرت دارای علائم بیماری قارچی موجود در سیلوهای دامداری و مرغداری­های مختلف استان خراسان رضوی انجام گرفت. با کشت دانه های ذرت بر روی محیط کشت های PDA و WA، پس از جداسازی و خالص سازی جدایه­های متعلق به جنس فوزاریوم، مجموعأ تعداد 40 جدایه بدست آمده که با توجه به ویژگی­های ماکروسکوپیک و میکروسکوپیک آنها و مقایسه با کلیدهای شناسایی مربوطه، سه گونه Fusarium verticillioides،   F. graminearum و F. proliferatum به ترتیب با فراوانی 34، 4 و 2 جدایه شناسایی شدند. به منظور تأیید نتایج بدست آمده، پس از استخراج DNA جدایه­ها و دانه­های ذرت با استفاده از واکنش PCR و آغازگرهای اختصاصی توالی مورد نظر تکثیر شد. بعلاوه، با استفاده از واکنش PCR به همراه آغازگر اختصاصی (FUM1 F/R) 15 جدایه از گونه­ی غالب F.verticillioides از نظر پتانسیل تولید فومونیزین مورد بررسی قرار گرفتند. در محصولات PCR، تولید نوارهای bp183 برای 15 جدایه مزبور مشاهده گردید. نتایج حاصل از این مطالعه نشان داد که همه­ی جدایه های مورد بررسی گونه­ی F. verticillioidesاز نظر ژنتیکی از پتانسیل تولید فومونیزین برخوردار هستند، همچنین با تکنیک مبتنی بر PCR و نشانگرهای مولکولی بکار رفته در این مطالعه، شناسایی، تفکیک  و ردیابی گونه­های فوزاریوم همراه دانه­ی ذرت مقدور می­باشد.

Abstract
Fusarium, a phytopathogenic fungus in fields of corn, not only reduces the quality and quantity of crops but also causes food contamination and endangers people  ̓s health by producing some mycotoxins such as Fumonisin (FUM). In the present study, samples were collected from fungal disease symptoms- showing corn from different livestock and poultry storage in Khorasan-Razavi province. Having placed the corn on PDA and WA, isolates belonging to Fusarium genus were isolated and purified. As a result, a total of 40 isolates were obtained among which three species were identified as Fusarium verticillioides, F. graminearum and F. proliferatum with isolated frequencies of 34, 4 and 2, respectively. To support these results,  DNA was extracted from the isolates from corn seeds and the fungal ribosomal DNA was amplified using PCR with species- specific primers. In addition, using PCR with the specific primers (FUM1 F/R) 15 isolates of F. verticillioides, as the prevalent species were examined for fumonisin production. The results indicated that all of the examined F. verticllioides isolates gave the expected fragment with  183 bp in size. Therefore, this study demonstrated that F. verticllioides isolates genetically have a potential for fumonisin-production. It also showed that the PCR-based technique can be used for the identification, distinction and detection of the Fusarium species.
