اثر سالیسیلیک اسید در القاء مقاومت سیستمیک در گیاه خیار به بیماری مرگ گیاهچه ناشی از Pythium aphanidermatum

چکیده
بیماری مرگ گیاهچه ناشی از(Edson) Fitzp. Pythium aphanidermatum یکی از بیماری­های مخرب خیار در گلخانه های تجاری است که موجب کاهش قابل توجهی از محصول می شود. در این پژوهش اثر اسید سالیسیلیک در القاء مقاومت به خیار آلوده به بیماری مرگ گیاهچه مورد بررسی قرار گرفت. بدین منظور، تغییر در سطح بیان ژن­های cupi4 و Lipoxygenase و فعالیت چند آنزیم بیوشیمیایی، مورد ارزیابی قرار گرفت. آزمایش به صورت فاکتوریل در قالب طرح بلوک کاملاً تصادفی با چهار تکرار در شرایط گلخانه در دمای 25 تا C˚ 27 با رطوبت نسبی 70% و دوره نوری 14:8 ساعت روشنایی: تاریکی انجام شد. نتایج ارزیابی وقوع بیماری نشان داد که بین شدت بیماری زایی در گیاهان تیمار شده با غلظت های مختلف اسید سالیسیلیک (صفر،۲۰۰ و۴۰۰ پی پی ام) و تیمار شاهد اختلاف معنی­داری در سطح احتمال 5% وجود دارد.  ارزیابی فعالیت آنزیم های آنتی  اکسیدانتی در گیاهان تیمار شده نشان داد که فعالیت  آنزیم های پراکسیداز و پلی فنل اکسیداز به میزان قابل ملاحظه­ای در مقایسه با شاهد افزایش یافت در حالیکه فعالیت آنزیم فنیل آلانین آمونیالیاز افزایش معنی­داری با گیاهان تیمار نشده نشان نداد. آنالیز بیان ژن با روش qRT-PCR نشان داد که کاربرد اسید سالیسیلیک قادر به افزایش سطح بیان ژن های بررسی شده می باشد اما بیشترین میزان بیان برای ژن Cupi4 ثبت گردید. نتایج این تحقیق نشان می­دهد که کاربرد اسید سالیسیلیک می­تواند نقش مهمی در افزایش مقاومت گیاه خیار در برابر بیماری مرگ گیاهچه ایفا  کند.
