شناسایی گونه های Pyrenophora، Bipolaris، Curvularia و Exserohilum مرتبط با بیماری‌های لکه برگی جو در استان آذربایجان شرقی با استفاده از داده های ریخت شناختی و مولکولی

چکیده
بیماری­ های برگی از مهمترین بیماری­ های جو در دنیا به ­شمار می­روند و باعث ایجاد خسارت اقتصادی در مناطق کشت و پرورش این محصول می­ شوند. تحقیق حاضر با هدف شناسایی گونه­ های Pyrenophora، Bipolaris، Curvularia و Exserohilum مرتبط با بیماری­های لکه ­برگی جو در استان آذربایجان­شرقی با استفاده از مفهوم تلفیقی گونه انجام گرفت. در مجموع 462 جدایه قارچی از گیاهان جو دارای علایم لکه برگی طی سال های 1396-1395 جداسازی و خالص ­سازی گردید. براساس تلفیق داده ­های ریخت ­شناختی با داده ­های توالی ناحیه ITS-rDNA و ژن گلیسرآلدهید-3- فسفات دهیدروژناز (gpdh) تنوع قابل توجهی در گونه ­های قارچی مرتبط با بیماری­های لکه­ برگی جو مشاهده گردید. در این تحقیق گونه­ های Pyrenophora graminea،P. teres ،Bipolaris sorokiniana ،B. salkadehensis،Curvularia inaequalis ، C. spicifera،C. nicotiae  وExserohilum rostratum  شناسایی گردیدند. دو گونه P. graminea وP. teres  به ترتیب با فراوانی 4/68 و 54/25 درصد، به عنوان گونه ­های قارچی غالب مرتبط با بیماری­ های لکه برگی جو بودند. با شناسایی گونه ­های قارچی مرتبط با این بیماری­ها، اعمال راهکارهای مناسب برای مدیریت آنها در این منطقه امکانپذیر خواهد شد.

Abstract
Foliar diseases are considered among important barely diseases worldwide, causing economic crop losses in barely industry worldwide, including Iran. Present study was aimed to characterize Pyrenophora, Bipolaris, Curvularia and Exserohilum species associated with leaf spot diseases of barley in East Azerbaijan province, using consolidated species concept. Totally 462 fungal isolates were isolated and purified from barley plants representing leaf spot symptoms during 2016-2017. Based on morphological and sequence data of ITS-rDNA and Glyceraldehyde-3-Phosphate Dehydogenase gene (gpdh) regions. A considerable diversity was observed among the fungal species associated with leaf spot diseases on barley. In this study, Pyrenophora graminea, P. teres, Bipolaris sorokinana, B. salkadehensis, Curvularia inaequalis, C. spicifera, C. nicotiae and Exserohilum rostratum were identified. Pyrenophora graminea and P. teres were dominant species associated with brown leaf spot symptoms, with isolation frequencies of 68.4 and 25.54 percent, respectively. With the identification of fungal species associated with leaf spot diseases on barley, it will be possible to practice suitable management strategies for these diseases.
