اثرات کشندگی آبامکتین و پروپارژیت بر کنه ی تارتن دو لکه ای Tetranychus urticae Koch (Acari, (Tetranychidae جمعیت مغان، و کارایی اثرات باقیمانده ی آنها در گیاه لوبیا
چکیده به منظور مقایسهی اثرات کشندهی آبامکتین و پروپارژیت و همچنین ماندگاری این اثرات روی گیاه لوبیا، افراد بالغ Tetranychus urticae در آزمایشهای جداگانه مورد تیمار قرار گرفتند. از روش آغشته کردن قطعهی برگی و قرار دادن آن روی بستر مرطوب، برای تیمار کنهها استفاده شد. پنج غلظت از هر ترکیب شیمیایی تهیه و برای هر غلظت در هر تکرار از 20 کنهی بالغ استفاده گردید و آزمایش حداقل سه بار تکرار شد واحد تکرار شامل یک پتریدیش با درپوش دارای تهویه و کفپوشی از پنبه مرطوب، حاوی قطعهی برگ (به طول و عرض دو سانتیمتر) آلوده به ترکیب شیمیایی بود که 20 کنهی بالغ روی آن قرار داشت. ثبت مرگ و میر 48 ساعت پس از تیمار انجام شد. برای بررسی کارایی باقیماندهی کنهکشها، گیاهان لوبیا با یک بیستم دوز توصیه شدهی مزرعهای تیمار شدند. در فواصل زمانی یک، سه، هفت، 10 ، 14 و 21 روز پس از تیمار، از برگها قطعههای برگی تهیه و 20 کنهی بالغ روی هر قطعه منتقل و پس از 48 ساعت تلفات ثبت گردید. آزمایش برای هر ترکیب سه بار تکرار شد و از آب مقطر به عنوان شاهد استفاده گردید. تجزیهی پروبیت دادههای آزمایش کشندگی نشان داد که مقادیر غلظت کشندهی 20 درصد، 50 درصد و 90 درصد برای آبامکتین به ترتیب 108/0، 417/0و 26/3 پیپیام و برای پروپارژیت به ترتیب 657/0، 26/2 و 90/14 پیپیام از فراوردهی تجاری بود. نتایج بررسی کارایی باقیماندهی کنهکشها روی گیاه لوبیا نشان داد که آبامکتین منجر به تلفات 100 درصد کنه در یک روز پس از سمپاشی با غلظت یک بیستم دوز توصیه شدهی مزرعهای گردید که این مقدار به 62/55 درصد در 21 روز پس از سمپاشی کاهش یافت. این مقدار برای پروپارژیت یک روز پس از سمپاشی 81 درصد بود که 21 روز پس از سمپاشی به صفر درصد کاهش یافت.

بررسی کاربرد جداگانه و تلفیقی قارچ i2375 Trichoderma harzianum و ورمی کمپوست در مهار زیستی نماتد ریشه ی گرهی (Meloidogyne javanica) گوجه فرنگی
چکیده در این تحقیق اثر تلفیقی و جداگانه قارچ i2375 Trichoderma harzianum و کود آلی ورمیکمپوست روی مهار بیماریزایی نماتد ریشهگرهی Meloidogyne javanica در شرایط گلخانهای ارزیابی گردید. با استفاده از ویژگیهای ریختشناسی، نماتد ریشهگرهی و جدایهی قارچ، مورد شناسایی قرار گرفتند. در شرایط گلخانهای اثر کاربرد تلفیقی و جداگانهی ورمیکمپوست و قارچ T. harzianum روی عوامل رشدی گیاه گوجهفرنگی و عوامل جمعیتی نماتد، دو بار مستقل از هم در دو زمان برداشت 90 و 120 روز در فصل بهار مورد بررسی قرار گرفتند. نتایج آزمایشات نشان داد که استفادهی همزمان ورمیکمپوست و قارچ تریکودرما، علاوه بر مهار نماتد ریشهی گرهی، عملکرد رشدی گیاه گوجهفرنگی از جمله طول و وزن تر اندام هوایی را افزایش داده که این نشان از توانایی مهار نماتد ریشهی گرهی با کاربرد تلفیقی دو عامل زیستی بوده است. تعداد گال در کاربرد تیمار تلفیقی دارای کمترین تعداد بوده و جمعیت نماتد در شرایط گلخانه تا 6/78 درصد توسط کاربرد همزمان قارچ و ورمیکمپوست مهار گردید، که تفاوت آنان با شاهد در سطح 5 درصد معنیدار بود. نوع تیمار اعمال شده با توجه به میزان کنترل نماتد نشان از نقش بسزای استفاده همزمان ورمیکمپوست و قارچ تریکودرما در کاهش شاخصهای آلودگی و جمعیتی نماتد داشته است.

بررسی تغییرات فعالیتهای آنزیمی در ریشه ی گیاه خربزه ی آلوده به عامل بیماری پژمردگی فوزاریومی (Fusarium oxysporum f. sp. melonis)
چکیده بیماری پژمردگی آوندی ناشی از Fusarium oxysporum f. sp. melonis یکی از بیماریهای مهم خربزه است. به منظور یافتن منبع مقاومت به نژاد 2.1 فوزاریوم، واکنش تعداد 18 توده خربزه جمعآوری شده از مناطق مختلف ایران در آزمایش گلخانهای در قالب طرح بلوکهای کامل تصادفی با سه تکرار در موسسهی تحقیقات اصلاح و تهیه نهال و بذر مورد ارزیابی قرار گرفتند. ریشههای گیاهچههای هر چهار توده در مرحله یک تا دو برگ حقیقی در 50 میلیلیتر سوسپانسیون اسپور Fusarium oxysporum f. sp. melonis با غلظت 106 اسپور در میلیلیتر به مدت 2-1 دقیقه قرار گرفتند و به سینیهای کشت برگردانده شدند. تودههای ایزابل مقاوم، سوسکی نیمه مقاوم، قصری حساس و شادگانی بسیار حساس جهت اندازهگیری ترکیبات فنل کل و تعیین میزان فعالیت آنزیمهای پراکسیداز، پلیفنل اکسیداز، کاتالاز و سوپراکسید دیسموتاز مورد استفاده قرار گرفتند. نمونهبرداریها از قسمت ریشه گیاهان در مراحل زمانی دو، چهار، شش و هشت روز بعد از مایهزنی برای بررسی تغییرات فعالیت آنزیمی و فنل کل انجام گرفت. بر اساس نتایج به دست آمده، میزان فعالیت آنزیمهای پراکسیداز، پلیفنل اکسیداز، کاتالاز و سوپراکسید دیسموتاز در طی روزهای مختلف در تودههای مختلف اختلاف معنیداری با یکدیگر داشتند. در روز چهارم و ششم حداکثر فعالیت آنزیم به ترتیب در تودههای مقاوم و بسیار حساس ایزابل و شادگانی ثبت شد. تغییرات ترکیبات فنلی در طی روزهای چهارم، ششم و هشتم با یکدیگر تفاوت معنیداری داشتند. در روز شاهد (روز صفر) بیشترین مقدار فعالیت در توده شادگانی (توده بسیار حساس) ثبت گردید در حالیکه پس از گسترش عامل بیماری بیشترین مقدار آن در روز چهارم پس از مایهزنی در توده مقاوم ایزابل بدست آمد. در این بررسی میزان فعالیت آنزیمهای پراکسیداز، پلیفنل اکسیداز، کاتالاز، سوپراکسید دیسموتاز و میزان ترکیبات فنلی به منزله پنج عامل اساسی دفاع بیوشیمایی در برهمکنش تودههای مختلف خربزه با عامل پژمردگی آوندی افزایش مییابند.

اثرات سه نوع کمپوست بر بیماری پژمردگی ورتیسیلیومی پنبه Verticillium dahliae در شرایط گلخانه
چکیده پنبه (Gossypium hirsutum) از گیاهان مهم صنعتی و استراتژیک بهشمار میرود که پژمردگی ورتیسیلیومی از مخربترین بیماریهای آن است. این تحقیق بهمنظور بررسی تأثیر سه نوع کمپوست (حاصل از ضایعات شهری، بقایای گیاهی و فراوردهی سیلوی دامپروری) بر پیشرفت بیماری پژمردگی ورتیسیلیومی پنبه رقم ورامین در گلخانه تحقیقاتی دانشگاه شهرکرد انجام گرفت. آزمایش بهصورت فاکتوریل در قالب طرح کاملاً تصادفی با چهار تکرار انجام شد. در تیمارهای مختلف، میکرواسکلروتهای Verticillium dahliae (پاتوتیپ غیربرگریز) به نسبتهای صفر، پنج و ده عدد در هر گرم خاک، و کمپوست مورد نظر به نسبت 35 درصد وزنی، قبل از کشت با خاک مخلوط شد. نتایج نشان داد که کمپوستهای بهکار رفته شاخص پژمردگی بوتهها را کاهش و وزن تر اندام هوایی را افزایش دادهاند. مقایسه میانگینها نشان داد که تیمار بقایای گیاهی و فراوردهی سیلو پژمردگی بوتهها را نسبت به شاهد کاهش معنیدار داده و در یک گروه آماری قرار گرفتند، درحالیکه تیمار کاربرد کمپوست ضایعات شهری از این نظر در گروه شاهد قرار گرفت. همچنین وزن تر اندام هوایی در کاربرد هر سه کمپوست افزایش معنیداری نسبت به شاهد نشان داد. برآورد جمعیت قارچ در خاک بیانگر کاهش بقای قارچ با کاربرد کمپوستها بود. بررسیهای آزمایشگاهی نشاندهندهی کاهش رشد ریسههای قارچ در محیط کشت حاوی عصاره کمپوست بقایای گیاهی بود. در مقایسهی سه کمپوست، اثر دو کمپوست بقایای گیاهی و فراوردهی سیلو بر شاخص پژمردگی، وزن تر اندام هوایی و جمعیت نهایی قارچ در خاک اختلاف آماری نشان نداد. بر اساس این نتایچ کاربرد دو کمپوست بقایای گیاهی و فراوردهی سیلو برای کاهش خسارت پژمردگی ورتیسیلیومی پنبه در قالب مدیریت تلفیقی بیماری مفید و مستلزم تحقیقات بیشتر است.

شناسایی گونه های تریکودرمای اندوفیت درختان بلوط در جنگل های ارسباران با استفاده از معیارهای ریخت شناختی و مولکولی
چکیده گونههای جنس Trichoderma از پراکنش جهانی برخوردار بوده و مایکوبیوتای غالب خاکهای مناطق مختلف میباشند. گونههای تریکودرما علیرغم اینکه از پتانسیل ساپروفیتی قوی برخوردار میباشند، دارای فاز اپیفیتکی قابل توجهی بوده و از پتانسیل نفوذ به بافتهای داخلی میزبان گیاهی بدون ایجاد خسارت و صدمه به میزبان برخوردار هستند. در سالهای اخیر، توجه ویژهای به شناسایی و برهمکنش گونههای تریکودرمای اندوفیت با میزبانهای گیاهی معطوف گردیده است، با این وجود، اطلاعات چندانی در مورد تنوع زیستی گونههای تریکودرمای اندوفیت در ایران وجود ندارد. تحقیق حاضر با هدف شناسایی گونههای تریکودرمای اندوفیت درختان بلوط در جنگلهای ارسباران انجام گردید. برای این منظور در تابستان سال 1393 از قسمتهای تنه و سرشاخهی درختان بلوط سالم و درختان دارای علایم زوال در مناطق حاتم بیگ )مشگین شهر) و کلیبر نمونهبرداری شد. در مجموع، 23 جدایهی تریکودرما از مناطق نمونهبرداری شده جداسازی و خالصسازی گردید. هویت جدایهها بر اساس ویژگیهای ریختشناختی و دادههای توالی ناحیه ITS-rDNA، Trichoderma atroviride، T. citrinoviride، T. harzianum، T. longibrachiatum و T. polysporum تعیین گردید. در تبارنمای ترسیم شده براساس توالی ناحیه ITS-rDNA، جدایههای مربوط به هر یک از گونههای شناسایی شده به همراه توالی نمونهی تیپ هریک از گونهها، با اعتبار سنجی بالا گروهبندی شدند. کلیهی گونهها برای اولین بار از جنگلهای ارسباران گزارش میشوند و بلوط سیاه (Quercus macranthera) به عنوان میزبان جدید برای تمامی گونههای شناسایی شده در این تحقیق معرفی می شود. گزارش این گونهها از روی بلوط سیاه برای دنیا جدید میباشد. با شناسایی این گونهها امکان ارزیابی کارایی این گونهها در کنترل زیستی بیمارگرهای گیاهی و تقویت رشد گیاهان فراهم خواهد شد.

تأثیر بیماری ویروسی ریزومانیا (Beet necrotic yellow vein virus)بر عملکرد و ویژگیهای کیفی چغندرقند
چکیده بیماری ریزومانیا در حال حاضر یکی از مهمترین بیماریهای چغندرقند در دنیا است. عامل این بیماری ویروس رگبرگ زرد نکروتیک چغندرقند(Beet necrotic yellow vein virus) است. این بیماری انتشار جهانی دارد. در مطالعهی حاضر تأثیر بیماری ریزومانیا بر عملکرد و ویژگیهای کیفی ارقام چغندرقند شامل عملکرد ریشه، درصد قند، عملکرد شکر، ضریب استحصال شکر و میزان ناخالصیهای غیرقندی (سدیم، پتاسیم و نیتروژن آمینه) بررسی شد. بدین منظور تعداد 12 رقم چغندرقند تک جوانه شامل شش رقم تجاری مقاوم به ریزومانیا، سه رقم متحمل و سه رقم حساس به بیماری در قالب طرح بلوکهای کامل تصادفی در سه تکرار تحت دو شرایط مزرعهی عاری از ریزومانیا و مزرعهی دارای آلودگی طبیعی به بیماری کشت شدند. این آزمایش بهمدت دو سال در ایستگاه مرکز تحقیقات و آموزش کشاورزی استان فارس (زرقان) انجام شد. نتایج نشان داد بیماری ریزومانیا با تأثیر منفی بر عملکرد ریشه و درصد قند باعث کاهش عملکرد شکر میشود. میانگین مقدار کاهش عملکرد شکر در اثر بیماری برای ارقام مقاوم تجاری، ارقام متحمل و ارقام حساس به ترتیب 25، 54 و 90% بود. در شرایط آلوده به ریزومانیا میزان سدیم در تمامی ارقام بهطور میانگین 165% افزایش یافت ولی میزان نیتروژن آمینه بهطور میانگین 43% کاهش نشان داد. تشکیل نشانههای بیماری بر روی ارقام مقاوم تجاری و نیز کاهش عملکرد آنها در شرایط آلوده به ریزومانیا نشان داد ژن Rz1 قادر به القاء مقاومت کامل در برابر بیماری نیست.

ارزیابی کارایی زنبور پارازیتویید Lysiphlebus fabarum (Marshall) (Hym., Braconidae) پرورشیافته روی شتهی سیاه باقلاAphis fabae Scopoli ، به منظور کنترل شتهی جالیز Aphis gossypii Glover
چکیده شتهی جالیز Aphis gossypii یکیاز آفات خسارت زای مهم در مزارع و گلخانههای صیفی و سبزی است. بررسی شاخصهای زیستی مرتبط با کارایی دشمنان طبیعی یکی از راهکارهای کنترل کیفی عوامل بیولوژیک قبل از رهاسازی محسوب میشود. در این مطالعه ویژگیهای زیستی (نرخ پارازیتیسم، نرخ ظهور و نسبت جنسی) جمعیت جنسی زنبور Lysiphlebus fabarum روی سنین مختلف رشدی شتهی جالیز بررسی شد. همچنین شاخصهای مهم در شایستگی نتاج (اندازهی بدن، طول دورهی رشدونمو، میزان باروری و طول عمر) مورد ارزیابی قرار گرفت. بر اساس نتایج بهدست آمده زنبور L. fabarum قادر بود تمام سنین پورگی شتهی جالیز را بهخوبی پارازیته کند. در تمام شاخصهای زیستی مورد بررسی بهجز اندازهی تخم، اختلاف معنیداری بین زنبورهای حاصل از چهار سن پورگی مشاهده شد به صورتی که رشد و نمو زنبور در پورههای سن دوم و سوم میزبان باعث تولید نتاج با ویژگیهای زیستی مطلوبتری نسبت به سایر مراحل رشدی شته شد. درمجموع نتایج پژوهش حاضر این نوید را میدهد که بتوان از این پارازیتویید در برنامههای کنترل بیولوژیک شتهی جالیز استفاده نمود؛ هرچند این نتایج باید در شرایط گلخانهای و مزرعهای نیز مورد ارزیابی قرار گیرد.

بررسی خصوصیات بیولوژیکی و مولکولی چند جدایه ویروس موزائیک خیارCucumber mosaic virus (CMV))) از مناطق شمال غرب ایران
چکیده ویروس موزائیک خیار یکی از مهمترین ویروسهای آلوده کنندهی گیاهی در ایران میباشد. به منظور بررسی فنوتیپ آلودگی جدایههای مختلف این ویروس در سال 1390 تعداد 342 نمونهی گیاهی مشکوک به آلودگی با این ویروس از گیاهان خیار، کدو، خربزه و گوجه فرنگی از مناطق مختلف شمال غرب کشور جمعآوری و با آزمون الایزا 91 نمونهی آلوده به CMV ردیابی شد. از بین جدایه ها هشت جدایه که توالی ژنومی و موقعیت تبارزایی آنها مشخص گردیده بود، برای مقایسهی خصوصیات بیولوژیکی و مطالعات بیشتر انتخاب شدند. شاخص بیماری برای این جدایه ها روی کدو محاسبه گردید. براساس علایم، جدایه ها در دو گروه متمایز شدند. بررسی بیان ژن پروتئین پوششی و سرکوب گر خاموشی RNA ((2b با استفاده از تکنیک RT-PCR نیمه کمی، نشان داد که غلظت جدایهها در گیاهان محک متفاوت میباشد. جدایههای ایرانی زیر گروه IB علائم شدیدتری را بر روی گیاهان محک نسبت به جدایه های زیر گروه IA نشان دادند. بنابراین به نظر میرسد که بین ویژگیهای ژنوتیپی و خصوصیات بیماری زایی جدایه ها ارتباط معنی داری وجود دارد.

کارآیی ژنهای مقاومت به بیماری لکهبرگی سپتوریایی(Stb) در ارقام افتراقی گندم در برابر جدایههای Zymoseptoria tritici
چکیده بیماری لکه برگی سپتوریایی گندم ناشی از Zymoseptoria tritici، از مهمترین بیماریهای گندم در جهان و ایران بهشمار میرود. مقاومت ژنتیکی مهمترین و اقتصادیترین راهبرد برای کنترل این بیماری میباشد. بنابراین پایش مداوم جمعیت قارچ بیمارگر برای بررسی اثربخشی ژن مقاومتStbدر برابر جدایههایZ. tritici ضروری است. در این پژوهش، الگوی پرآزاری پنج جدایهی مختلف روی ارقام افتراقی گندم حاوی ژنهای مقاومت و اثر بخشی این ژنها در برابر جدایهها در مرحلهی گیاهچهای و در شرایط گلخانه مورد بررسی قرار گرفت. نتایج نشان دادکهجدایههای BK94 و BK95 پرآزارترین جدایه بودند که روی تعداد بیشتری از ارقام افتراقی بیماریزایی داشتند و در مقابل، جدایهی BK56 کمترین پرآزاری را روی ارقام افتراقی نشان داد. ارقام M3 و Arina در برابر تمام جدایههای مورد بررسی مقاوم بودند. ارقام Riband، TE9111 وFlame بهترتیب در برابر چهار، سه و دو جدایهی مورد بررسی مقاومت نشان دادند. بقیهی ارقام افتراقی در برابر جدایههای مورد بررسی حساس بودند.ژنهایStb15، Stb16 و Stb17 موثرترین ژنهای مقاومت در برابر همهی جدایهها بودند، بنابراین ارقام محتوی این ژنها، میتوانند بهعنوان منابع مقاومت موثر در برنامه های اصلاح ارقام جهت مقاومت در برابر لکه برگی سپتوریایی گندم مورد استفاده قرار گیرند.

ارزیابی اثر علفکشهای انتخابی سویا بر سرعت رشد میسلیوم قارچ Macrophomina phaseolina (Tassi) Goid عامل پوسیدگی ذغالی در شرایط آزمایشگاهی
چکیده علفکشها بهطور وسیعی در مزارع مختلف مورد استفاده قرار میگیرند اما مکانیسم فعل و انفعلات ممکن بین علفکشها و بیمارگرهای گیاهی بهخوبی شناخته نشده است. یکی از مهمترین بیماریهای سویا، پوسیدگی ذغالی است که عامل آن قارچ Macrophomina phaseolina ، میباشد. علفکشهای ایمازتاپیر، تریفلورالین و متریبیوزین به صورت کاربرد خاکی در کشت سویا کاربرد دارند. بهمنظور بررسی اثر غلظتهای مختلف علفکشهای مذکور بر سرعت رشد قارچ، آزمایشی بهصورت فاکتوریل در قالب طرح کاملاً تصادفی در شش تکرار در شرایط آزمایشگاهی انجام گردید. برای هر یک از علفکشها، غلظتهای صفر تا 7000 میلیگرم در لیتر در محیط کشت سیبزمینی دکستروز آگار (س. د. آ.) در نظر گرفته شد. دیسک قارچ در مرکز پتریدیشهای نه سانتیمتری قرار داده شد و میزان رشد طولی میسلیوم بهطور روزانه اندازهگیری گردید. میزان سرعت رشد با استفاده از معادله دز- پاسخ لجستیک چهار پارامتره برازش داده شد. نتایج حاصل از تأثیر استفاده از غلظتهای علفکش در محیط کشت نشان داد که بین جدایهها تفاوت معنیداری وجود دارد. علفکشهای تریفلورالین با غلظتهای 4320 و 5760 میلیگرم در لیتر، ایمازتاپیر در غلظتهای 2300 و 7000 میلیگرم در لیتر و همچنین متریبیوزین در غلظتهای 5000 و 7000 میلیگرم در لیتر تقریباً به طور کامل موجب توقف رشد قارچ شدند. علفکشهای ایمازتاپیر، متریبیوزین و تریفلورالین بهترتیب بیشترین تأثیر را روی کاهش سرعت رشد جدایههای M. phaseolina در محیط کشت س. د. آ. داشتند.

اثر تغذیه ی لاروها از گلیسرول و آب نوشیدن حشرات کامل روی چند ویژگی زیستی شب پره ی مدیترانهای آرد، Ephestia kuehniella Zeller (Lep.; Pyralidae)
چکیده
تغذیهی حشرات کامل شبپرههای انباری جنس Ephestia از آب و محلولهای قندی، و نیز کیفیت و کمیت رژیم غذایی لاروها بسیاری از ویژگیهای زیستی حشرات کامل را تحت تاثیر قرار میدهند. در این تحقیق، اثر تغذیهی لاروهای شبپرهی مدیترانهای آرد (Ephestia kuehniella Zeller) از گلیسرول و آب نوشیدن حشرات کامل بر طول عمر و زادآوری حشرات کامل بررسی شد. لاروها روی دو رژیم غذایی: جوانهی گندم، مخمر آبجو و گلیسرول (10 به 1 به 2)، و جوانهی گندم و مخمر آبجو (10 به 1) پرورش داده شدند. برخی حشرات کامل پیش از جفتگیری به مدت دو روز آب نوشیدند و برخی به آب دسترسی نداشتند. اثر تغذیهی لاروها از گلیسرول در صورت آب ننوشیدن حشرات کامل نر و ماده روی طول عمر مادهها معنیدار بود ولی آب نوشیدن یک و یا هر دو جنس باعث عدم تاثیر آن شد. آب نوشیدن هر دو جنس، بسته به تغذیه یا عدم تغذیهی لاروها از گلیسرول، به ترتیب باعث کاهش معنیدار و غیرمعنیدار طول عمر مادهها شد. اثر تغذیهی لاروها از گلیسرول روی طول عمر نرها نیز معنیدار بود ولی برخلاف مادهها، آب نوشیدن نرها نبود گلیسرول را جبران نکرد. حشرات کامل حاصل از رژیم غذایی دارای گلیسرول، زادآوری بالاتری داشتند. آب نوشیدن یا ننوشیدن یک جنس، آب نوشیدن جنس دیگر را روی زادآوری تحت تاثیر قرار داد. تغذیه از گلیسرول درصد تفریخ تخمها را افزایش داد و اثر جفتگیری نرها یا مادههای آب نوشیده یا ننوشیده با هم، روی درصد تفریخ تخمها متفاوت بود. به طور کلی، گلیسرول رژیم غذایی و آب نوشیدن حشرات کامل باعث افزایش طول عمر و زادآوری آنها شد و درصد تفریخ تخمها تحت تاثیر گلیسرول رژیم غذایی و آب نوشیدن حشرات کامل نر قرار داشت.

چکیده ویروس موزاییک رگهای گندم (WSMV) Wheat streak mosaic virus از ویروسهای خسارت زای غلات مربوط به خانواده Potyviridae و جنس Tritimovirus است. در این تحقیق، NIa-Pro که یکی از مهمترین پروتئینازهای این ویروس میباشد، مورد مطالعه قرار گرفت. بدین منظور 116 نمونهی علائمدار گندم از مزارع خراسان شمالی، خراسان جنوبی و آذربایجان غربی جمعآوری و به منظورشناسایی اولیه، کلیهی نمونهها با آزمون الایزا آزمایش گردیدند و تعداد 10 نمونه در مقابل آنتی بادی اختصاصی WSMV واکنش مثبت نشان دادند. این 10 نمونه مثبت در گلخانه روی گیاه گندم تکثیر و با آغازگر اختصاصی مربوط به توالی کامل NIa-Pro در آزمون RT-PCR سنجش شده و قطعهای به طول 732 جفت باز تکثیر گردید. سپس دو جدایه متفاوت از نظر جغرافیایی مربوط به آذربایجان غربی و خراسان شمالی، همسانه سازی و تعیین ترادف شد. در مقایسهی ترادفهای به دست آمده در این تحقیق با ترادف های موجود در ژن بانک، دو گروه تشکیل شد. جدایههای ایرانی مربوط به این مطالعه در گروه یک، زیر گروه IB و مجزا از جدایههای اروپایی و جدایه ایرانی که در تحقیقات پیشین تعیین توالی شده بود و نزدیک به جدایه آمریکایی (NC-001886)، بودند قرار گرفتند. بررسی مکانهای برشی در ژن NIa-Pro نشان داد که مناطق برش، در این پروتئین حفاظت شده است، جایگزینی تعدادی اسید آمینه در مکانهای حفاظت شده نظیر توالیGKSH در محل 124-121، A/T در جایگاه 154و گلوتامین با آلانین در جایگاه 208 در جدایههای ایرانی مشهود بود. تحقیق حاضر اولین بررسی جدایههای ایرانی در ناحیهی کد کننده مذکور میباشد.
