شناسایی و بررسی بیماریزایی گونه های قارچی همراه با بیماری لکه برگی .Cucumis sativus L خیار در استان آذربایجان شرقی
چکیده
بیماری لکهبرگی بعنوان یکی از بیماریهای محدودکنندهی تولید محصول خیار در استان آذربایجانشرقی شناخته شده است و همه ساله خسارت زیادی به این محصول و جالیزکاران وارد مینماید.هدف تحقیق حاضر شناسائی عامل بیماری لکهبرگی خیار Cucumis sativus L.در استان آذربایجانشرقی و شناسایی گونههای قارچی بیمارگر با استفاده از دادههای مورفولوژیکی میباشد. برای این منظور در طی تابستان سال 1392 از مزارع خیار دارای علایم در شهرستانهای شبستر، اهر، میانه و باسمنج که از مراکز مهم تولید خیار در استان هستند، نمونهبرداری به عمل آمد و از تعداد 90 نمونه گیاهی تهیه شده که دارای علائم متنوع لکه برگی بودند، تعداد 100 جدایهی قارچی جدا سازی گردید. نتایج این تحقیق حضور انواع گونههای مختلف قارچی عامل لکهبرگی را در مزارع استان تایید میکند. با مطالعات ریختشناختی جدایه های بدست آمده، گونههای قارچیAlternaria tenuissima ،Bipolaris spicifera ،Bipolaris sorkiniana ،Stemphylium mali ،Alternaria atrum ،Cladosporium sp.،Nigrospora oryzae ،Pseudoprenospora cubensis ، Phoma sp.وCurvularia sp. . به عنوان عوامل ایجاد لکه برگی خیار جداسازی شدند. دربین گونههای شناسایی شده، فراوانی جدایههایAlternaria tenuissima و Pseudoprenospora cubensisبترتیب با 48 درصد و13 درصد بیشتر از بقیه بود. تمام جدایه های مورد بررسی قادر به ایجاد بیماری در بوته های خیار مایه زنی شده بودند..

مروری بر آلودگی نماتد مولد گره ریشه در باغ های پسته ی رفسنجان
چکیده به منظور بررسی وضعیت آلودگی باغهای پسته (Pistacia vera L.) به نماتد مولد گره ریشه (M. incognita)، نمونههای خاک و ریشهی درختان و علفهای هرز از مناطق پستهکاری شهرستان رفسنجان طی ماههای مختلف در سالهای 92-1391 جمعآوری شد. با استفاده از خصوصیات ریخت شناسی مادههای بالغ و لاروهای سن دوم نماتد، گونهی غالب M. incognita تشخیص داده شد. بررسی پویایی جمعیت لارو سن دوم نماتد در خاک نشان داد که کمترین دامنهی جمعیتی به ترتیب در ماههای خرداد، مهر، آبان و اسفند با تعداد پنج تا هفت و حداکثر آن با تعداد 50 تا 210 نماتد در 100 گرم خاک در مرداد ماه متغیر بود. بالاترین تراکم جمعیت با تعداد 98 گال و 53 نماتد ماده در هر گرم ریشه در ماه مرداد مشاهده شد. جمعیت نماتد گره ریشه از پنج تا 300 لارو سن دوم در 100 گرم خاک به ترتیب در ماههای خرداد و مرداد متغیر بود. 18 گونه گیاهی به عنوان میزبان گونهی M. incognitaبه ثبت رسیدند در حالیکه گونهی M. javanica تنها روی پنج گونه گیاهی مشاهده شد. نتایج این تحقیق میتواند در تعیین زمان دقیق مبارزه با نماتد ریشهگرهی و مدیریت علفهای هرز در باغهای پسته مفید واقع گردد.

تعیین ترادف ژن پروتئین پوششی دو جدایه ایرانی ویروس موزائیک هندوانه و بررسی جایگاه فیلوژنتیکی آنها
چکیده
ویروس موزاییک هندوانه (WMV) Watermelon mosaic virusمتعلق به جنس Potyvirus و خانواده Potyviridae از ویروسهای مهم آلوده کننده کدوئیان در دنیا محسوب میشود. دو جدایه این ویروس به نامهای YAZ.WMV.41 و YAZ.WMV.55 از مزارع خیار و طالبی استان یزد جمعآوری گردید. آلودگی نمونهها با آزمون سرولوژیکی الایزای مستقیم و غیرمستقیم و آنتی بادی پلیکلونال این ویروس بررسی شد. این دو جدایه در مقابل آنتیبادی واکنش نشان ندادند. در بررسی علائم این جدایهها برروی گیاهان آزمون کدوی مراغه، لوبیا چیتی و لوبیا چشم بلبلی، علائم متفاوتی با آنچه تا کنون در مورد جدایههای این ویروس گزارش شده، مشاهده گردید. با استفاده از آغازگرهای اختصاصی قطعهای به طول 809 جفت باز از ژن پروتئین پوششی تکثیر و تعیین ترادف شد. مقایسهی ترادفهای بدست آمده در این تحقیق با ترادفهای موجود در بانک جهانی ژن نشان داد که جدایههای مورد مطالعه به همراه یک جدایهی ایرانی (رس شمار EU667627) با دامنهی میزبانی متفاوت، در زیر گروه IIB قرار میگیرند. سایر جدایههای ایرانی گزارش شده قبلی در گروههای I و AII قرار گرفتند. مقایسهی میزان تشابه اسید نوکلئیک این جدایهها با یکدیگر نشان داد که جدایههای ایرانی مربوط به این مطالعه بیشترین و کمترین میزان شباهت را با جدایههای کشورهای اسپانیا (رس شمار AJ579518) و ایران (رس شمار GQ421161) به ترتیب به میزان 5/99 و 2/92 درصد دارا میباشند. نتایج حاصل از این تحقیق بیانگر این است که کدوئیاندر مزارع وگلخانههای ایران بوسیله جدایههای متعدد این ویروس آلوده می شوند.

شیوع گونههای Penicillium روی انگور و کشمش در استانهای آذربایجانشرقی، غربی و قزوین
چکیده
اعضای جنس پنیسیلیوم محصولات کشاورزی را در طی مراحل قبل و بعد از برداشت آلوده نموده و باعث کاهش کیفیت و کمیت محصول میگردند. اطلاعات زیادی در مورد شیوع گونههای این جنس در روی انگور و کشمش در ایران وجود ندارد. در این تحقیق شیوع گونههای پنیسیلیوم همراه انگور و کشمش در تاکستانهای استانهای آذربایجانشرقی، غربی و قزوین مورد بررسی قرار گرفت. طی فصلهای رویشی 1392-1390 تعداد 63 جدایه متعلق به جنس پنیسیلیوم از روی انگور و کشمش در استانهای مورد مطالعه جداسازی گردید. میانگین درصد کل آلودگی 23% و میانگین آلودگی در نمونههای کشمش و انگور بهترتیب 1% و 33% بود. بررسی ویژگیهای ریختشناختی جدایهها براساس کلیدهای شناسایی گونههای پنیسیلیوم، جدایههای پنیسیلیوم همراه انگور و کشمش در این منطقه را در نه گونهی پنیسیلیوم و تالارومیسس قرار داد. هویت گونهها با استفاده از دادههای توالی ژن بتاتوبولین تایید گردید. نتایج این بررسی نشان داد که گونهیPenicillium expansumبا فراوانی 35% (22 جدایه) بیشترین تعداد جدایهها را به خود اختصاص داد. شناسایی گونههای پنیسیلیوم همراه انگور و کشمش در تاکستانهای منطقه شمالغرب ایران میتواند راهکار مناسب مدیریت در تولید محصول عاری از آلودگی را فراهم نماید.

رابطه بین جمعیت شتههای ناقل با انتشار ویروسها در سیبزمینی بذری: مطالعه موردی در منطقه فریدن اصفهان
چکیده
شتهها یکی از آفات مهم سیبزمینی هستند که خسارت اصلی آنها مربوط به انتقال بیماریهای ویروسی است. در این مطالعه تراکم جمعیت شتههای ناقل، درصد بیماریهای ویروسی و رابطه بین آنها در یک مزرعه آزمایشی که در آن غدههای عاری از ویروس به مدت دو سال کشت شده بود، بررسی گردید. از کارتهای چسبدار زرد رنگ جهت شکار شتهها استفاده شد و در آزمایشگاه تراکم تجمعی جمعیت شتهها در دو گروه شته سبز هلو Myzus persicae (به عنوان مهمترین گونه غالب) و سایر شتهها تعیین گردید. رابطه بین تراکم جمعیت شتهها (در دو گروه شته سبز هلو و سایر شتهها)، درصد وقوع بیماریهای ویروسی مختلف شاملPVY،PLRV، PVM و PVS و رابطهی بین آنها با محاسبهی شیب رگرسیون خطی و ضریب همبستگی ارزیابی شد. رابطهی بسیار معنیداری بین تراکم شتهی سبز هلو با تراکم سایر شتهها وجود دارد. همچنین رابطهی بین ویروسهای مختلف بجز PLRV با تراکم جمعیت شتهی سبز هلو و تراکم سایر شتهها معنیدار بود. این نتایج نشان میدهد که علیرغم فعالیت شتهی سبز هلو و اهمیت زیاد آن در انتقال ویروسهای مختلف در سیبزمینی بذری، در شرایط منطقهی فریدن سایر گونهها نیز در انتقال ویروسهای مورد نظر دخالت دارند. وجود رابطهی بسیار معنیدار بین PVY با شتههای ناقل، انتشار بیشتر این ویروس را در مزارع بذری منطقه توجیه مینماید.

حساسیت چندگونه آفت انباری به اسانس های گیاهان شوید، کرفس و نعناع وحشی
چکیده
استفاده از آفتکشهای شیمیایی علاوه بر تهدید سلامتی انسان، خطرات محیطی را هم بهوجود آورده است. اسانسهای گیاهان معطر بهعنوان حشرهکشهای طبیعی و سالم برای کنترل حشرات آفت مطرح شدهاند. در تحقیق حاضر، سمیت اسانس گیاهان شوید، کرفس و نعناع وحشی روی حشرات کامل سوسک چهارنقطهای حبوبات (Callosobruchus maculatus F.) ، لاروهای شبپرهی مدیترانهای آرد (Ephestia kuehniella Zeller) و شبپرهی هندی (Plodia interpunctella (Hübner)) بررسی شد. بر اساس نتایج تجزیهی شیمیایی اسانسها با دستگاه گاز کروماتوگرافی-طیفسنج جرمی، (+)-کاروون (81/48 درصد)، آپیول (58/22 درصد) و لیمونن (61/12 درصد) در اسانس شوید، آپیول (82/21 درصد)، دی-لیمونن (21/21 درصد)، 3-کارن (80/11 درصد) در اسانس کرفس و میریستیسین (35/13 درصد)، 3-ترپینولنون (33/13 درصد) و 2-ایزوپروپیلدن سیکلوهگزانون (239/9 درصد) دراسانس نعناع وحشی بهعنوان ترکیبهای اصلی شناسایی شدند. اسانسها سمیت تدخینی بالایی را روی هر سه گونه آقت نشان دادند. LC50اسانسهای نعناع وحشی، شوید و کرفس روی حشرات کامل سوسک چهارنقطهای حبوبات بهترتیب 958/58، 769/67 و 888/94 میکرولیتر بر لیتر برآورد شدند. لاروهای شبپرهی هندی نسبت به لاروهای شبپرهی مدیترانهای آرد و حشراتکامل سوسک چهارنقطهای حبوبات در برابر اسانس شوید حساستر بودند. اسانس نعناع وحشی سمیت تدخینی بیشتری نسبت به اسانسهای شوید و کرفس روی شبپرهی مدیترانهای آرد نشان داد. همچنین لاروهای شبپرهی هندی نسبت به حشرات کامل سوسک چهارنقطهای حبوبات حساسیت بیشتری در برابر اسانس نعناع وحشی نشان دادند. نتایج این پژوهش نشان داد که اسانسهای شوید، کرفس و نعناعوحشی دارای پتانسیل حشرهکشی مناسبی بوده و میتوانند در مدیریت سوسک چهارنقطهای حبوبات، شبپرهی مدیترانهای و شبپرهی هندی مد نظر قرار گیرند.

بررسی بیمارگری قارچ Metarhizium anisopliae بر روی شته سبز هلو Myzus persicae
چکیده یکی از مشهورترین قارچهای بیمارگر حشرات، anisopliae Metarhiziumمی باشد که در خاک وجود داشته و از قابلیت بیمار کردن آفات گیاهی و جانوری برخوردار میباشد. شته ی سبز هلو(Myzus persicae) در کلیهی مناطق ایران وجود داشته و به درختان هلو، مرکبات، سبزیجات و گیاهان زینتی گلخانهای خسارت می زند. در این مطالعه مرگ و میر شتهی سبز هلو توسط قارچ Metarhiziumanisopliae مورد بر رسی قرار گرفت. از دو جدایه Metarhizium anisopliae به نام های A3 و M1 موجود در گروه میکروبیولوژی سازمان جهاد دانشگاهی تهران استفاده شد. تست زیست سنجی علیه شته ی سبز هلو در داخل ظروف پتری 18 سانتیمتری با غلظتهای 103× 1، 105×1 و 107×1 کنیدی در میلی لیتر در شرایط 14:10 ساعت تاریکی: روشنایی و دمای 28 درجهی سانتیگراد به مدت 9 روز انجام گرفت. کمترین مقدار LC50 برای جدایه A3 غلظت 105×5/6 کنیدی در میلیلیتر بود و بهترین LT50 نیز برای جدایه A3 برابر با 82/3 روز در غلظت 107×1 کنیدی در میلیلیتر بود. کمترین مقدار LT50 جدایهی M1 نیز در غلظت 107×1 برابر با 51/5 روز بود.

تأثیر عصاره ی پنج گونهی گیاهی روی لاروهای سن دوم سوسک کلرادوی سیب زمینی Leptinotarsa decemlineata (Say) (Coleoptera: Chrysomelidae)
سوسک کلرادو یکی از مهمترین آفات حشرهای سیبزمینی میباشد. استفادهی مکرر از آفتکشهای شیمیایی برای کنترل این آفت باعث آلودگی محیط زیست، مقاوم شدن آفت به آفتکشها و نابودی دشمنان طبیعی شده است. برخی عصارههای گیاهی با تجزیهپذیری و ایمنی بالای خود میتوانند جایگزین مناسبی برای آفتکشهای شیمیایی در مدیریت تلفیقی این آفت باشند. در این پژوهش تأثیر عصارهی برگهای درخت گردو Juglans regia L. ، آقطی Sambucus ebulus L.، گندواش Artemisia annua L.، مرزهی سهندی Satureja sahendica Bornm و مریم گلی سهندی Salvia sahendica Boiss and Buhse روی لاروهای سن دوم سوسک کلرادوی سیبزمینی Leptinotarsa decemlineata (Say) در شرایط آزمایشگاهی (دمای 2± 26 درجهی سانتیگراد، رطوبت نسبی 5 ± 55 درصد و دورهی نوری 16 ساعت روشنایی و 8 ساعت تاریکی) بررسی شد. زیستسنجیها به روش غوطهورسازی برگهای سیبزمینی در غلظتهای 625/0، 25/1، 5/2، 5 و 10 درصد عصارهی گیاهان مذکور و شاهد متانول و آب مقطر انجام شدند. برای هر غلظت سه تکرار در نظر گرفته شد و در هر تکرار 20 لارو سن دوم تا 24 ساعته استفاده شد. مرگ و میر لاروها، 24، 48، 72 و 96 ساعت پس از تیمار ثبت شد. نتایج نشان دادند که عصارهی سه گیاه گردو، مرزهی سهندی و گندواش در مقایسه با آقطی و مریم گلی سهندی در غلظتهای مختلف، تأثیر بیشتری روی لاروهای سن دوم داشتند. به طوری که 96 ساعت بعد از تیمار، در غلظت 10% مرگ و میر ناشی از عصارههای مذکور بهترتیب به 33/93، 67/91 و 33/73% و در غلظت 5% به ترتیب به 9/58، 100 و 2/48 درصد رسید. با توجه به درصد تلفات قابل توجه بهویژه در غلظتهای بالا، عصارههای این گیاهان میتوانند به عنوان یک منبع گیاهی بالقوه در برنامهی مدیریت تلفیقی سوسک کلرادوی سیبزمینی کاربرد داشته باشند.

اثر ارقام مختلف تجاری بادام روی واکنش های زیستی شتهی آردی بادامHyalopterus amygdali Blanch.(Hom., Aphididae) در شرایط مزرعه
چکیده استان آذربایجانشرقی یکی از مهمترین مراکز عمدهی تولیدکنندهی محصول بادام و نهال اصلاح شدهی این گونه گیاهی با ارزش در کشور است. درختان بادام در طول زندگی خود مورد حملهی آفات متنوعی از جمله شتهها قرار میگیرند. شتهی آردیHyalopterus amygdali (Blanchard, 1840) گونهی غالب در روی درختان بادام در استان آذربایجان شرقی است. این آفت با تغذیه از شیرهی گیاهی برگ و سرشاخهی بادام خسارت قابل توجهی وارد میسازد. مطالعهی حاضر به منظور تشخیص برخی از ویژگیهای آنتیبیوزی چند رقم تجاری بادام با نامهای سهند (بومی منطقه)، شکوفه، آذر (ارقام اصلاح شده داخلی)، فرانیس و نوپلوساولترا (ارقام خارجی) روی شتهی آردی بادام تحت شرایط مزرعهای انجام شد. صفات مورد اندازهگیری شامل تعیین درصد بقای پورهها، طول دورهی نشو و نمای پورگی، شاخص رشد، قدرت باروری و نرخ ذاتی افزایش جمعیت (rm) حشرات کامل بیبال شته اردی بادام بود. تجزیه واریانس دادهها نشان داد که بین ارقام مورد مطالعه بادام از نظر تأثیر روی طول دورهی نشو و نمای پورگی و قدرت باروری شتهی آردی بادام اختلاف معنیداری وجود نداشته ولی اختلاف بین میانگین نرخ ذاتی افزایش جمعیت (rm value) حشرات کامل بیبال در سطح احتمال پنج درصد معنیدار است. مطابق این یافتهها، میانگین کل میزان بقای پورهها 86/3 ± 68 درصد، طول دورهی نشو و نمای پورگی 11/0 ± 96/8 روز، تعداد نتاج تولید شده به ازای یک فرد ماده 21/2 ± 02/38 پوره و نرخ ذاتی افزایش جمعیت برابر با 0043/0 ± 2581/0 ماده/ماده/روز به دست آمد. مقایسهی گروهی ارقام اصلاح شده با رقم بومی منتخب از تودههای منطقه (سهند) حاکی از وجود اختلاف معنیدار بین رقم سهند با ارقام آذر، شکوفه، فرانیس و نوپلوساولترا (ارقام اصلاح شده) در تمامی صفات مورد مطالعه در سطح احتمال یک درصد بود. شتههای پرورش یافته روی رقم سهند دارای باروری کمتر و نرخ ذاتی افزایش جمعیت پایینتر بود. بنابراین میتوان اظهار نمود که سهند در مقایسه با سایر ارقام به طور نسبی یک رقم مقاوم به شتهی آردی بادام است.
