اثر چند ترکیب مختلف روی فعالیت آنزیم آلفا- آمیلاز روده میانی شبپره مدیترانهای آرد
Anagasta kuehniella (Zeller, 1879) (Lep., Pyralidae)
در اینبررسی اثر هفت نوع ترکیب مختلف روی فعالیت آنزیم آلفا- آمیلاز روده میانی لاروهایسن پنجم افراد نر و ماده شبپره مدیترانهای آرد مطالعه گردید. میزان فعالیت ویژه آنزیمبا استفاده از کیت تشخیص فعالیت آلفا- آمیلاز اندازهگیری شد. غلظت پروتئین کل درنمونههای آنزیمی به روش بردفورد تعیین گردید. تأثیر درجات مختلف اتانول (صفر، 25،50، 75، 85 و 96 درصد) و غلظتهای صفر، 1، 2، 3، 4 و 5 میلیمولار NaCl، EDTA، Tris، SDSنیترات منیزیم و فسفات پتاسیم روی فعالیت آنزیم درلاروهای نر و ماده بررسی شد. میانگینهای مربوط به اثر اتانول روی فعالیت آنزیم دارایاختلاف معنیدار بودند و بیشترین میزان مهار آنزیم توسط الکل 96 درصد صورت گرفت.فعالیت آنزیم در غلظتهای یک تا پنجمیلیمولار NaCl در لاروهای هر دو جنس دارای اختلاف معنیدار بودو با افزایش غلظت NaCl میزان فعالیت آنزیم کاهش یافت.با افزایش غلظت SDS فعالیت آنزیم کاهش پیدا کرد طوریکه بیشترین فعالیت آنزیم لاروهاینر و ماده در غلظت یک میلیمولار و کمترین فعالیت در غلظت پنج میلیمولار مشاهده شد. اثر غلظتهای مختلفEDTA روی فعالیت آنزیم در لاروهای ماده دارای اختلاف معنیدار بود ولیدر لاروهای نر اختلاف معنیداری مشاهده نگردید. اثر غلظتهای مختلف Tris روی فعالیت آنزیم در هر دو جنس دارای اختلاف معنیدار بود و باافزایش غلظت از فعالیت آنزیم کاسته شد. فعالیت آنزیم در غلظتهای مختلف نیتراتمنیزیم اختلاف معنیداری را نشان داد و با افزایش غلظت، میزان فعالیت آنزیم کاهشپیدا کرد. فعالیت آنزیم در غلظتهای مختلف فسفات پتاسیم در لاروهای نر و مادهدارای اختلاف معنیدار بود و با افزایش غلظت از یک به پنج میلیمولار، فعالیت آنزیم افزایش پیدا نمود.آگاهی از چگونگی تأثیر مواد مختلف روی فعالیت آنزیمهای آلفا- آمیلاز میتواند درامر شناسایی و تهیه مهارکنندههای آنزیمها برای کنترل با آفات موثر باشد.

معرفی بخشی از فون کنه¬های میان¬استیگما (Acari: Mesostigmata) روی درخت¬چه¬های تمشک (Rubus spp.) در حوزه مرکزی استان گیلان
در طول فصول مختلف سال 1388، 230 نمونه در حوزه مرکزی استان گیلان جمعآوری شد. در نتیجه، تعداد 5323 کنه میاناستیگمای فعال روی درختچههای تمشک نقاط مختلف بهدست آمد. بر اساس نتایج، 33 گونه متعلق به 18 جنس و هفت خانواده از این راسته تشخیص داده شد. درمیان گونههای شناساییشده 12 گونه برای فون کنههای استان گیلان و دو گونه برای فون کنههای ایران جدید میباشند که بهترتیب با یک و دو ستاره مشخص شدهاند. فراوانی کنههای میاناستیگما در مقایسه با سایر راستههای کنهها بیشتر بود. گونههای جمعآوریشده بر حسب خانواده شامل:

بهمنظور تعیین مشخصات مولکولی و بررسی ساختارهای ثانویه ویروئید لکه زردی- 1 مو، 137 نمونه از تاکستانهای استانهای اردبیل، آذربایجان شرقی و آذربایجان غربی جمعآوری گردید. اسید نوکلئیک کل از بافت برگ نمونهها استخراج و پس از ساخت دیانای مکمل با استفاده از آغازگر ششنوکلئوتیدی تصادفی، طول کامل این ویروئید با روش پیسیآر و با استفاده از آغازگرهای اختصاصی ویروئید لکه زردی -1 مو، تکثیر شد. محصول پیسیآر پس از همسانهسازی در حامل pGEMT-easy، تعیینتوالی گردید. نتایج نشان داد که واریانتهای شناساییشده در این تحقیق، دارای 367 یا 368 نوکلئوتید هستند و در مجموع هفت واریانت جدید از این ویروئید تشخیص داده شد. درخت تبارزایی نشان داد که واریانت 6 مشابه اعضای تیپ یک این ویروئید و شش واریانت بعدی در کنار تعدادی جدایه و واریانت دیگر تشکیل یک تیپ جداگانه (بهنام پیشنهادی تیپ 4) را میدهند. در ساختار ثانویه واریانتهای بهدستآمده در این تحقیق، بیشترین تفاوت در منطقه متغیر و قسمت انتهایی چپ قرار داشت. ویروئید لکه زردی-1 مو برای اولین بار از استانهای آذربایجانشرقی و اردبیل گزارش میشود.

کارایی نماتد بیمارگر حشرات Heterorhabditis bacteriophora در کنترل زیستی نماتد گره ریشه Meloidogyne javanica در شرایط آزمایشگاهی
در سالهای اخیر، کنترل زیستی نماتدهای بیمارگر گیاهی با استفاده از نماتدهای بیمارگر حشرات مورد توجه قرار گرفته است. این تحقیق تاثیر لاروهای آلودهکننده زنده و مرده نماتد بیمارگر حشرات Heterorhabditis bacteriophora Poinar در غلظتهای 5، 10، 25، 50 و 100 لارو در هر میلیلیتر را بهطور جداگانه و در ظرفهای 24 چاهکی روی مرگ ومیر لارو وتفریخ تخم نماتد گره ریشه Meloidogyne javanica Treub در غلظتهای 50 تخم، 50 لارو و مخلوط 50 تخم و لارو به ازای هر چاهک در شرایط آزمایشگاهی مورد بررسی قرار داد. آزمایش با شش تیمار در قالب طرح کرتهای خردشده در زمان و چهار تکرار برای هر تیمار انجام شد. کل آزمایشها دو بار تکرار گردید. یادداشتبرداری نتایج بعد از 48، 72، 96 و120 ساعت نشان داد که تاثیر لارو زنده یا مرده نماتد بیمارگر حشرات در مرگومیر لاروهای سن دوم و تفریخ تخم نماتد گره ریشه معنیدار است. بیشترین مرگومیر در غلظت100 لارو آلودهکننده در هر میلیلیتر و در 120ساعت و بیشترین بازدارندگی از تفریخ تخم با غلظت 100 لارو آلودهکننده در هر میلیلیتر و در 48 ساعت مشاهده شد. در حالت مخلوط 50 تخم و لارو نماتد گره ریشه، تاثیر لاروهای زنده H. bacteriophora در مرگومیر لارو نماتد گره ریشه در بین زمانها اختلاف معنیداری بروز نداد.

توانایی اکتینومیست¬ها در کنترل زیستی برخی قارچ¬های بیمارگر و نماتد مرکبات در شرایط آزمایشگاهی
نماتد مرکبات، Tylenchulus semipenetrans، دارای گسترش جهانی بوده و یکی از عوامل سرخشکیدگی در باغهای مرکبات محسوب میشود. آلودگی توأم نماتد و قارچهای بیمارگر، تأثیر مضاعفی بر رشد گیاه میگذارد. بهمنظور بررسی اثر اکتینومیستها در کنترل نماتد و بیمارگرهای ثانویه، تعداد 30 نمونه خاک آلوده از فراریشه مرکبات شرق استان گیلان و غرب استان مازندران جمعآوری شدند. در نتیجه20 جدایه اکتینومیست با استفاده از کشت روی محیطهای انتخابی جداسازی و اثر آنها در تفریخ تخم و مرگومیر لاروها مورد ارزیابی قرار گرفت. در آزمایشی دیگر، تاثیر اکتینومیست ها بر قارچهای بیماریزای Fusarium oxysporum، Fusarium solani، Penicilium digitatum ، Rhizoctonia solani،Pestaliopsis spp و Colletotrichum gloeosporioides بررسی شد. در آزمون اول، هشت جدایه با توان کاهش میزان تفریخ تخم در مدت هفت روز و کشتن لاروها در طول چهار روز، از قابلیت آنتاگونیستی خوبی برخوردار بودند. در آزمون دوم، شش جدایه موفق به کنترل قارچهای بیمارگر موردمطالعه شدند. جدایه Streptomyces sp. IGM05 بیشترین توانایی را در کنترل قارچهای بیمارگر نشان داد در حالیکه جدایه Streptomyces sp. IGM17 با کاهش تفریخ تخم به میزان 2/37 درصد و مرگومیر 4/52 درصدی لاروها، توانایی آنتاگونیستی بالایی در برابر نماتد بروز داد.

شناسایی ریخت¬شناختی و مولکولی عوامل قارچی همراه با پوسیدگی میوه زیتون در استان زنجان
پوسیدگی میوه یکی از مهمترین بیماریهای زیتون در استان زنجان بهشمار میرود. علیرغم شیوع این بیماری در منطقه، عوامل قارچی مرتبط با این بیماری ناشناخته باقی ماندهاند. این تحقیق با هدف شناسایی گونههای قارچی عوامل پوسیدگی میوه زیتون در استان زنجان با استفاده از دادههای ریختشناختی و مولکولی انجام شد. برای این منظور از باغهای زیتون منطقه طارم در استان طی ماههای مهر و آبان سال 1390 نمونهبرداری صورت گرفت. پس از جداسازی قارچهای عامل بیماری، خالصسازی به روش تکاسپور و نوکریسه انجام شد. شناسایی جدایههای قارچی در سطح گونه با بررسیهای ریختشناختی و دادههای توالی نوکلئوتیدی ناحیه ITS-rDNA صورت گرفت. نتایج این بررسی نشان داد گونه مرکبAlternaria alternata با فراوانی 37 درصد بهعنوان گونه قارچی غالب در ایجاد بیماری پوسیدگی میوه در استان زنجان می باشد. در مورد جدایههای متعلق به جنس آلترناریا هویت گونه با استفاده از دادههای توالی نوکلئوتیدی ناحیه ITS-rDNA و توالی ژنهایgpd وrpb2 مورد تایید قرار گرفت، تمامی جدایهها در خوشه گونه مرکبAlternaria alternata قرار گرفتند. در درخت تبارزایی ترسیمشده بر اساس توالی rpb2 برای این جنس، جدایههای زیتون همراه با جدایههای گونه مرکب A. alternata یک خوشه را به خود اختصاص دادند. دیگر گونههای قارچی جداسازی شده از میوههای زیتون با علایم پوسیدگی این منطقه شامل Acremonium implicatum، Arthrinium phaeospermum،Aspergillus ochraceus، Aureobasidium pullulans،Botrytis cinerea ، Epicoccum nigrum، Fusarium sp.، F. proliferatum، F. nygamai،Penicillium expansum، Trichoderma harzianumوTruncatella angustata بودند. گزارش اغلب گونههای قارچی از روی زیتون برای ایران جدید میباشد. با شناسایی گونههای قارچی همراه با بیماری پوسیدگی میوه زیتون، اعمال راهکارهای مناسب برای مدیریت بیماری در آینده امکانپذیر خواهد شد.

شناسایی گروه¬های سازگاری رویشی و دودمان¬های کلونال قارچ Fusarium solani عامل پوسیدگی ریشه لوبیا در زنجان
گروههای سازگاری رویشی 42 جدایهFusarium solani عامل بیماری پوسیدگی ریشه لوبیا جمعآوریشده از مناطق مختلف لوبیاکاری استان زنجان، با استفاده از جهشیافتهگان nit تعیین شدند. فنوتیپ جهشیافتهگان nit بر اساس مشخصات رشدی آنها در محیط غذایی حداقل (MM) حاوی منابع مختلف نیتروژن تعیین گردید و جهشیافته nitM بیشترین فراوانی جهشیافتهها را بهخود اختصاص داد. آزمون مکملسازی و تعیین گروههای سازگاری رویشی برای جدایههای خودسازگار صورت گرفت. بر این اساس، جدایهها در 16 گروه VCG قرار گرفتند که چهار گروه از آنها تکعضوی و بقیه چندعضوی بودند. گروههای چندعضوی همچنین دربرگیرنده جدایههای متصلکننده یا پل بودند که بهطور همزمان در بیش از یک گروه قرار گرفتند. ارتباطی بین گروههای سازگاری رویشی و منطقه جغرافیایی مشاهده نگردید. جهت تعیین دودمانهای کلونال، هشت آغازگر تصادفی مورد استفاده قرار گرفتند و از بین آنها یک آغازگر (OPA-13) با تولید باندهای تکرارپذیر و چندشکلی مناسب انتخاب شد. واکنش زنجیرهای پلیمراز از بین 42 جدایه برای 26 جدایه، موفق بود و نتایج مولکولی تنوع بالای این جدایهها را نشان داد بهطوریکه آنها را در 13 دودمان کلونالی با ضریب تشابه 75 درصد گروهبندی نمود. هر دودمان کلونالی متشکل از تنها یک هاپلوتیپ یک یا چندعضوی بود. بین گروهبندی بر اساس روش مولکولی و سازگاری رویشی همبستگی بارزی مشاهده نگردید.

شته مومی کلم یکی از آفات مهم کلزا است. بهمنظور ارزیابی مقاومت 21 ژنوتیپ کلزا در برابر شته مومی کلم طی سالهای 87-1385، آزمایشی در شرایط مزرعهای و گلخانهای انجام گرفت. در هر سال دو آزمایش جداگانه و هر کدام در قالب طرح بلوکهای کامل تصادفی با سه تکرار در شرایط آلودگی طبیعی و عدمآلودگی در مزرعه اجرا گردید. شاخص آلودگی و شاخص کاهش تابعی عملکرد در مزرعه محاسبه شد. بهمنظور مقایسه میزان تحمل گلخانهای، ژنوتیپها در قالب طرح کاملاً تصادفی در دو آزمایش مجزای آلوده و عدمآلوده، هر کدام با سه تکرار در گلدان کاشته شدند و شاخصهای کاهش تابعی طول ساقه و خسارت واردشده به گیاهان اندازهگیری گردیدند. در آزمایش آنتیبیوز، نرخ ذاتی افزایش جمعیت شته مومی کلم روی ژنوتیپها در قالب طرح کاملاً تصادفی در 10 تکرار در شرایط گلخانهای محاسبه شدند. نتایج تجزیه واریانس مرکب کرتهای خردشده در زمان نشان داد که شاخص آلودگی در بین ژنوتیپها، سالهای اجرای آزمایش و زمانهای نمونهبرداری اختلاف معنیداری داشتند. ژنوتیپ Opera با 23/4 کمترین شاخص آلودگی را داشت. تجزیه مرکب شاخص کاهش عملکرد دانه و اجزای آن اختلاف معنیداری را بین ژنوتیپها نشان داد. شاخصهای خسارت و کاهش طول ساقه ژنوتیپها اختلاف معنیداری در آزمایش تحمل گلخانه داشتند. در بین ژنوتیپها از نظر نرخ ذاتی افزایش جمعیت شته در آزمایش گلخانهای آنتیبیوز، تفاوت معنیدار مشاهده شد. ژنوتیپOkapi با 21/0rm= (ماده/ماده/روز) کمترین مقدار نرخ ذاتی افزایش جمعیت را داشت. در نهایت، ژنوتیپهای Okapi و Opera به ترتیب با 58/23 و 78/19 بیشترین مقادیر شاخص مقاومت گیاهی را نسبت به شته مومی کلم نشان دادند.
