Despite the incidence of the disease in the region, the fungal species involved in occurrence of the disease largely remain unknown. The aim of present study is to characterize fungal species associated with olive fruit rot in Zanjan province based on morphological and molecular data. For this purpose, olive fruit samples showing fruit rot symptoms were collected from olive orchards in Zanjan province, during the September and October 2011. Pure cultures were established using a single spore technique. Fungal isolates were identified using morphological characteristics as well as sequence data of ITS-rDNA region. The results revealed Alternaria alternata species complex as the dominant species amongst the fungal groups involved in olive fruit rot in this region with a frequency of 37 percent. The identity of Alternaria isolates were further analysed based on sequence data from the rpb2 and gpd genes. The phylogeny inferred using sequence data from rpb2 gene culstred olive isolates in A. alternata species complex clade. The other fungal species identified in this study include Acremonium implicatum, Arthrinium phaeospermum, Aspergillus ochraceus, Aureobasidium pullulans, Botrytis cinerea, Epicoccum nigrum, Fusarium sp., F. nygamai, F. proliferatum, Penicillium expansum, Trichoderma harzianum and Truncatella angustata. Most of the species represent new records for Iran. With the identification of fungal species associated with olive fruit rot, it will be possible to practice suitable management strategies for this disease.}, keywords = {fruit rot,ITS-rDNA,Phylogeny,Alternaria,Fusarium,RPB2}, title_fa = {شناسایی ریخت¬شناختی و مولکولی عوامل قارچی همراه با پوسیدگی میوه زیتون در استان زنجان}, abstract_fa = {پوسیدگی میوه یکی از مهم­ترین بیماری­های زیتون در استان زنجان به­شمار می­رود. علی­رغم شیوع این بیماری در منطقه، عوامل قارچی مرتبط با این بیماری ناشناخته باقی مانده­اند. این تحقیق با هدف شناسایی گونه­های قارچی عوامل پوسیدگی میوه زیتون در استان زنجان با استفاده از داده­های ریخت­شناختی و مولکولی انجام شد. برای این منظور از باغ­های زیتون منطقه طارم در استان طی ماه­های مهر و آبان سال 1390 نمونه­برداری صورت گرفت. پس از جداسازی قارچ­های عامل بیماری، خالص­سازی به روش تک­اسپور و نوک­ریسه انجام شد. شناسایی جدایه­های قارچی در سطح گونه با بررسی­های ریخت­شناختی و داده­های توالی نوکلئوتیدی ناحیه ITS-rDNA صورت گرفت. نتایج این بررسی نشان داد گونه مرکبAlternaria alternata  با فراوانی 37 درصد به­عنوان گونه قارچی غالب در ایجاد بیماری پوسیدگی میوه در استان زنجان می باشد. در مورد جدایه­های متعلق به جنس آلترناریا هویت گونه با استفاده از داده­های توالی نوکلئوتیدی ناحیه ITS-rDNA و توالی ژن­هایgpd  وrpb2  مورد تایید قرار گرفت، تمامی جدایه­ها در خوشه گونه مرکبAlternaria alternata  قرار گرفتند. در درخت تبارزایی ترسیم­شده بر اساس توالی rpb2 برای این جنس، جدایه­های زیتون همراه با جدایه­های گونه مرکب A. alternata یک خوشه را به خود اختصاص دادند. دیگر گونه­های قارچی جداسازی شده از میوه­های زیتون با علایم پوسیدگی این منطقه شامل Acremonium implicatum، Arthrinium phaeospermum،Aspergillus ochraceus، Aureobasidium pullulans،Botrytis cinerea ، Epicoccum nigrum، Fusarium sp.، F. proliferatum، F. nygamai،Penicillium expansum، Trichoderma harzianumوTruncatella angustata  بودند. گزارش اغلب گونه­های قارچی از روی زیتون برای ایران جدید می­باشد. با شناسایی گونه­های قارچی همراه با بیماری پوسیدگی میوه زیتون، اعمال راه­کارهای مناسب برای مدیریت بیماری در آینده امکان­پذیر خواهد شد.}, keywords_fa = {پوسیدگی میوه,تبارزایی,Alternaria,Fusarium,ITS-rDNA,RPB2}, url = {https://arpp.tabrizu.ac.ir/article_585.html}, eprint = {https://arpp.tabrizu.ac.ir/article_585_1fb02719171f65c07fde553af241b6ef.pdf} }
